How does the production of Romeo and Juliet designed and directed by Éric Ruf, the administrator of the Comédie-Française since 2014, figure out William Shakespeare’s tragedy in an unexpected way? And how does it transform the tragedy into a tragi-comedy? These are questions that the following article tries to answer. The first and major part of the study focuses on Éric Ruf’s parti pris concerning the myth that Romeo and Juliet has become over time. What is the stage-director’s position vis-à-vis the Shakespearean text and what is the interpretation he shares with his audience? What is the gap between the original script – here in François-Victor Hugo’s translation – and the audio-visual adaptations of the play? Furthermore, how does Juliette (Suliane Brahim) seem to be the heroine of a family vendetta while Roméo (Jérémy Lopez) has more of the anti-hero? Finally how does Éric Ruf compose the mythic balcony scene and thwart the stereotypes about it? The last part of the article deals with the music partition that is omnipresent in the performance. Background: Most melanoma patients with BRAFV600E positive tumors respond well to a combination of BRAF kinase and MEK inhibitors. However, some patients are intrinsically resistant while the majority of patients eventually develop drug resistance to the treatment. For patients insufficiently responding to BRAF and MEK inhibitors, there is an ongoing need for new treatment targets. Cellular metabolism is such a promising new target line: mutant BRAFV600E has been shown to affect the metabolism. Methods: Time course experiments and a series of western blots were performed in a panel of BRAFV600E and BRAFWT/ NRASmut human melanoma cells, which were incubated with BRAF and MEK1 kinase inhibitors. siRNA approaches were used to investigate the metabolic players involved. Reactive oxygen species (ROS) were measured by confocal microscopy and AZD7545, an inhibitor targeting PDKs (pyruvate dehydrogenase kinase) was tested. Results: We show that inhibition of the RAS/RAF/MEK/ERK pathway induces phosphorylation of the pyruvate dehydrogenase PDH-E1α subunit in BRAFV600E and in BRAFWT/NRASmut harboring cells. Inhibition of BRAF, MEK1 and siRNA knock-down of ERK1/2 mediated phosphorylation of PDH. siRNA-mediated knock-down of all PDKs or the use of DCA (a pan-PDK inhibitor) abolished PDH-E1α phosphorylation. BRAF inhibitor treatment also induced the upregulation of ROS, concomitantly with the induction of PDH phosphorylation. Suppression of ROS by MitoQ suppressed PDH-E1α phosphorylation, strongly suggesting that ROS mediate the activation of PDKs. Interestingly, the inhibition of PDK1 with AZD7545 specifically suppressed growth of BRAF-mutant and BRAF inhibitor resistant melanoma cells. Conclusions: In BRAFV600E and BRAFWT/NRASmut melanoma cells, the increased production of ROS upon inhibition of the RAS/RAF/MEK/ERK pathway, is responsible for activating PDKs, which in turn phosphorylate and inactivate PDH. As part of a possible salvage pathway, the tricarboxylic acid cycle is inhibited leading to reduced oxidative metabolism and reduced ROS levels. We show that inhibition of PDKs by AZD7545 leads to growth suppression of BRAF-mutated and -inhibitor resistant melanoma cells. Thus small molecule PDK inhibitors such as AZD7545, might be promising drugs for combination treatment in melanoma patients with activating RAS/RAF/MEK/ERK pathway mutations (50% BRAF, 25% NRASmut, 11.9% NF1mut). Abstract—In this paper we propose ROS-Defender, a holistic approach to secure robotics systems, which integrates a Security Event Management System (SIEM), an intrusion prevention system (IPS) and a firewall for a robotic system. ROS-Defender combines anomaly detection systems at application (ROS) level and network level, with dynamic policy enforcement points using software defined networking (SDN) to provide protection against a large class of attacks. Although SIEMs, IPS, and firewall have been previously used to secure computer networks, ROSDefender is applying them for the specific use case of robotic systems, where security is in many cases an afterthought. Reactive Oxygen Species (ROS) generation is an unavoidable background process in the normal functioning of the cell. The greatest contributor to ROS production is the electron transport chain (ETC) where O2 is reduced to H2O. Some incompletely-reduced oxygen species escape and oxidize a variety of organic molecules (e.g. proteins and lipids in the mitochondrial membrane), leading to molecular dysfunction and initiating a positive feedback loop leading to generation of even more active radicals. Increased ROS concentration damages mitochondria and further increases ROS generation. Healthy cells manage ROS enzymatically with superoxide dismutase and other enzymes, various antioxidants, and ultimately through increased mitophagy of damaged mitochondria. The precise tuning of the latter mechanism is crucial for cell survival and is controlled in the cell by a ROS-induced regulatory network, which consists of many components such as Nrf2, Keap1, Parkin and p62 with a rather complicated cross-talk (Figure 1). In many diseases (cancer, Parkinson’s disease (PD), Huntington’s disease (HD), etc.), various components of the ROS management network are altered. Deconstructing the molecular mechanisms underlying or resulting from these alterations might contribute to better understanding of the dynamics of related pathophysiological processes. We have built a kinetics-based model which recapitulates the consensus understanding of the mechanism responsible for cellular ROS – managing system. [less ▲]

Nitrate tolerance is associated with an enhanced superoxide anion (O(2)(-)) production and may be attenuated by statins as they interact with the two main endothelial NO synthase (eNOS) and NAD(P)H oxidase pathways involved in this oxidative stress. Groups of wild-type (wt, C57Bl/6J) and eNOS knock-out mice (eNOS(-/-)) received rosuvastatin (20 mg kg(-1) day(-1) p.o.) for 5 weeks and a cotreatment with the statin plus nitroglycerin (NTG; 30 mg kg(-1) day(-1), subcutaneous injections b.i.d.) for the last 4 days. Another group received only NTG (30 mg kg(-1) d(-1), b.i.d. for 4 days) and finally control mice from both strains received no treatment. Rings of thoracic aortas from these groups were studied in organ baths. Relaxations to NTG (0.1 nM-0.1 mM) were determined on thromboxane analogue (U44619)-precontracted rings and O(2)(-) production (RLU 5 s(-1) mg(-1) of total protein content) was assessed in aorta homogenates with the lucigenin-enhanced chemiluminescence technique. Reverse transcriptase-polymerase chain reaction analysis was performed on aortas from both mice strains. In vivo NTG treatment induced a significant rightward shift of the concentration-effect curve to NTG compared to control group. There was, however, no cross-tolerance with non-nitrate sources of NO (unaltered response to acetylcholine in wt group). The rosuvastatin + NTG cotreatment was able to protect against the development of nitrate tolerance in both mice strains and L-mevalonate abolished this protective effect of rosuvastatin. In vivo treatment with apocynin, a purported NAD(P)H oxidase inhibitor, also produced a similar protection to that observed with rosuvastatin in both strains.Superoxide anion formation was increased after NTG treatment in both mice strains and the rosuvastatin + NTG cotreatment was able to reduce that production. Moreover, rosuvastatin treatment abolished the increase in gp91phox mRNA (an endothelial membrane NAD(P)H oxidase subunit) expression induced by in vivo exposure to NTG. These findings suggest that long-term rosuvastatin treatment protects against nitrate tolerance by counteracting NTG-induced increase in O(2)(-) production, probably via a direct interaction with the NAD(P)H oxidase pathway.
