BACKGROUND: Stress-induced hypertrophic remodeling is a critical pathogenetic process leading to heart failure. Although many signal transduction cascades are demonstrated as important regulators to facilitate the induction of cardiac hypertrophy, the signaling pathways for suppressing hypertrophic remodeling remain largely unexplored. In this study, we identified p21-activated kinase 1 (Pak1) as a novel signaling regulator that antagonizes cardiac hypertrophy. METHODS AND RESULTS: Hypertrophic stress applied to primary neonatal rat cardiomyocytes (NRCMs) or murine hearts caused the activation of Pak1. Analysis of NRCMs expressing constitutively active Pak1 or in which Pak1 was silenced disclosed that Pak1 played an antihypertrophic role. To investigate the in vivo role of Pak1 in the heart, we generated mice with a cardiomyocyte-specific deletion of Pak1 (Pak1(cko)). When subjected to 2 weeks of pressure overload, Pak1(cko) mice developed greater cardiac hypertrophy with attendant blunting of JNK activation compared with controls, and these knockout mice underwent the transition into heart failure when prolonged stress was applied. Chronic angiotensin II infusion also caused increased cardiac hypertrophy in Pak1(cko) mice. Moreover, we discovered that the Pak1 activator FTY720, a sphingosine-like analog, was able to prevent pressure overload-induced hypertrophy in wild-type mice without compromising their cardiac functions. Meanwhile, FTY720 failed to exert such an effect on Pak1(cko) mice, suggesting that the antihypertrophic effect of FTY720 likely acts through Pak1 activation. CONCLUSIONS: These results, for the first time, establish Pak1 as a novel antihypertrophic regulator and suggest that it may be a potential therapeutic target for the treatment of cardiac hypertrophy and heart failure. We propose the use of password-authenticated key exchange (PAKE) for achieving and enhancing entity authentication (EA) and key management (KM) in the context of decentralized end-to-end encrypted email and secure messaging, i.e., without a public key infrastructure or a trusted third party. This not only simplifies the EA process by requiring users to share only a low-entropy secret such as a memorable word, but it also allows us to establish a high-entropy secret key. This approach enables a series of cryptographic enhancements and security properties, which are hard to achieve using out-of-band (OOB) authentication. We first study a few vulnerabilities in voice-based OOB authentication, in particular a combinatorial attack against lazy users, which we analyze in the context of a secure email solution. We then propose tackling public key authentication by solving the problem of secure equality test using PAKE and discuss various protocols and their properties. This method enables the automation of important KM tasks such as key renewal and future key pair authentications, reduces the impact of human errors and lends itself to the asynchronous nature of email and modern messaging. It also provides cryptographic enhancements including multi-device synchronization, and secure secret storage/retrieval, and paves the path for forward secrecy, deniability and post-quantum security.We also discuss the use of auditable PAKEs for mitigating a class of online guess and abort attacks in authentication protocols. We present an implementation of our proposal, called PakeMail, to demonstrate the feasibility of the core idea and discuss some of its cryptographic details, implemented features and efficiency aspects. We conclude with some design and security considerations, followed by future lines of work. [less ▲]

Palmerston English is spoken by just over 50 people on a remote atoll of the Cook Islands in the South Pacific. In 1863, the Englishman William Ma(r)sters settled on Palmerston with several Polynesian women from the Northern Group of the Cook Islands. Marsters ruled the island in an autocratic style: only English was allowed to be spoken and it is recorded that the children from his various wives spoke English with a strong British accent. The perpetuation of the speech was more difficult for the women than for the men because they did not have a direct female example from Britain to imitate. The three branches of the family are now in their 8th generation and their offspring live mainly in Rarotonga, Aitutaki, New Zealand and Australia (several thousand people, with a varying degree of competence in the language, depending on the contact retained with Palmerston). HIP14 is the most highly conserved of 23 human palmitoyl acyltransferases (PATs) that catalyze the post-translational addition of palmitate to proteins, including huntingtin (HTT). HIP14 is dysfunctional in the presence of mutant HTT (mHTT), the causative gene for Huntington disease (HD), and we hypothesize that reduced palmitoylation of HTT and other HIP14 substrates contributes to the pathogenesis of the disease. Here we describe the yeast two-hybrid (Y2H) interactors of HIP14 in the first comprehensive study of interactors of a mammalian PAT. Unexpectedly, we discovered a highly significant overlap between HIP14 interactors and 370 published interactors of HTT, 4-fold greater than for control proteins (P = 8 × 10(-5)). Nearly half of the 36 shared interactors are already implicated in HD, supporting a direct link between HIP14 and the disease. The HIP14 Y2H interaction set is significantly enriched for palmitoylated proteins that are candidate substrates. We confirmed that three of them, GPM6A, and the Sprouty domain-containing proteins SPRED1 and SPRED3, are indeed palmitoylated by HIP14; the first enzyme known to palmitoylate these proteins. These novel substrates functions might be affected by reduced palmitoylation in HD. We also show that the vesicular cargo adapter optineurin, an established HTT-binding protein, co-immunoprecipitates with HIP14 but is not palmitoylated. mHTT leads to mislocalization of optineurin and aberrant cargo trafficking. Therefore, it is possible that optineurin regulates trafficking of HIP14 to its substrates. Taken together, our data raise the possibility that defective palmitoylation by HIP14 might be an important mechanism that contributes to the pathogenesis of HD. [less ▲]

Comme le genre qu’il désigne, le terme est avant tout polémique : l’assimilation d’un écrit à un pamphlet, péjorative ou non, n’en est pas moins révélatrice des tensions à l’oeuvre dans le lexique politique de la Restauration. Comment celle-ci entend-elle le terme de « pamphlet » ? Comment s’illustre-t-il dans les champs politique et littéraire ?
