The present paper sketches the three main options - i.e. servicing EU clients directly from the UK, creating a branch or establishing a subsidiary - comparing their benefits and restrictions under the GATS rules and the free movement of capital and freedom of establishment under the EU Treaties. The focus is on traditional banking activities leaving aside investment services. Whilst acting through a EU based subsidiary appears to be the most promising option for UK banks who ambition to serve clients on a EU scale, the choice of location is not trivial as Member States are competing to attract such subsidiaries to their jurisdiction. The ECB in its supervisory role of the banking sector has already signaled that it will not allow such competition to result in a supervisory arbitrage. We propose an alternative that is based on the ESP (empirical saddlepoint) approximation of the solutions to the empirical moment conditions. Saddlepoint approximations are known to perform well in small sample. The novel estimator proposed, which we call the ESP estimator, is the mode of the ESP approximation. We show that it is consistent and asymptotically normal, and we study its higher-order bias. We propose novel test statistics based on the ESP estimator. Finally, we also investigate the finite-sample properties of the ESP estimator and related test statistics through Monte-Carlo simulations. [more ▼] La 1e Guerre mondiale fut le catalyseur de mutations sociales et techniques, qui suscitent de nouveaux besoins d’administration, matérialisés par l’émergence d’un nouveau groupe social: celui des experts. Dans les États modernes allemands et polonais, un groupe particulier d’experts se détache, qui se spécialisent dans une thématique aux profondes racines culturelles : l’Est européen.Les changements de régime et de territoire entre 1918 et 1972 déstabilisent le cadre de référence des sociétés allemandes et polonaises, particulièrement entre 1939 et 1945. Cependant, l’historiographie des sciences de l’Est allemandes (Ostforschung) souligne les continuités personnelles, institutionnelles et conceptuelles fortes dans l’expertise de l’Est du « IIIe Reich » à l’Allemagne de l’Ouest. En Allemagne de l’Est et en Pologne, au contraire, le changement de régime après 1945 rend impossible toute continuité, mais la question de l’évolution de la pensée sur l’Est dans les cercles de l’expertise, qui eux se maintiennent, reste posée.En dépassant l’analyse de l’expertise en termes de subordination aux différents régimes politiques auxquels elle s’adresse, nous mettons en valeur les spécificités de l’expertise de l’Est. Elle se caractérise par un double ancrage dans l’imaginaire collectif et dans les pratiques scientifiques contemporaines, pour exprimer un but politique. Cet ancrage explique l’inertie dans les conceptions allemandes et polonaises de l’Est. We work with ASPIC-END, a recently proposed adaptation of the structured argumentation framework ASPIC+ which can incorporate debates about logical principles, natural deduction style arguments and explanations. We apply this framework to build a preliminary formal model of parts of the debate that mathematicians had about the Axiom of Choice in the early 20th century. Furthermore, we briefly discuss the insight into the strengths and drawbacks of the modeling capacities of ASPIC-END that we have gained from producing this model. [more ▼] The cytoskeleton is an important system in health and disease. Composed by three major classes of filaments, it ensures the maintaining of the cellular shape, it is involved in cell motility and cell division. The complex regulation of this system is mediated by a various number of proteins which often display multiple and differential functions. Testin is one of these cytoskeletal proteins described in important processes involving the actin cytoskeleton such as cell migration and cell spreading. This thesis is dedicated to the study of testin during the cell division process which is mainly regulated by the actin and microtubule cytoskeleton. Our results provided evidence that, during mitosis, testin is able to dynamically localize to mitotic structures like centrosome, midzone and midbody. Further investigations of this localization, particularly in cytokinesis, permit to conclude that testin is a midbody bulge protein capable to concentrate to this intercellular bridge through its LIM1 and LIM2 domains. Testin is involved in the regulation of the actin cytoskeleton through being part of a protein complex with amongst other zyxin and talin, also able to concentrate to the midbody. Therefore, using RNAi experiments, we showed that the localization of testin to the midbody is zyxin and talin independent. In order to understand the potential function of testin during the cell division process, we initiate the identification of the potential partners of testin during mitosis. Based on the known interactors of testin, themselves described to localize to the midbody, we performed pull down as well as knock-down experiments. We also started the screening of potential mitotic partners of testin using the BioID approach. Given these observations about the localization of testin during mitosis the question arose whether testin plays a role in this process of cell division. Indeed, modulating the expression of testin results in different phenotypes. Overexpression of testin leads in faulty spindle formation or in a lack of abscission thus ultimately to an increase in multinucleated cells. Conversely, down regulation of testin leads to a perturbation of the nuclear shape and the microtubule structure of the midbody. Taken together, these data suggest that, in addition to its important role in actin-based processes in interphase, testin, a novel midbody bulge protein is involved in mitosis where it appears required for the integrity of a microtubule structure. [more ▼] Upon stimulation of cells with transforming growth factor beta (TGF-beta), Smad proteins form trimeric complexes and activate a broad spectrum of target genes. It remains unresolved which of the possible Smad complexes are formed in cellular contexts and how these contribute to gene expression. By combining quantitative mass spectrometry with a computational selection strategy, we predict and provide experimental evidence for the three most relevant Smad complexes in the mouse hepatoma cell line Hepa1-6. Utilizing dynamic pathway modeling, we specify the contribution of each Smad complex to the expression of representative Smad target genes, and show that these contributions are conserved in human hepatoma cell lines and primary hepatocytes. We predict, based on gene expression data of patient samples, increased amounts of Smad2/3/4 proteins and Smad2 phosphorylation as hallmarks of hepatocellular carcinoma and experimentally verify this prediction. Our findings demonstrate that modeling approaches can disentangle the complexity of transcription factor complex formation and its impact on gene expression.
